Telencephalon?specific <i>Alkbh1</i> conditional knockout mice display hippocampal atrophy and impaired learning

چکیده
AlkB homolog 1 (ALKBH1) is responsible for the biogenesis of 5-formylcytidine (f5C) on mitochondrial tRNAMet and essential protein synthesis. The brain, especially hippocampus, highly susceptible to dysfunction; hence, maintenance activity strongly required prevent disorders associated with hippocampal malfunction. To study role ALKBH1 in we generated dorsal telencephalon-specific Alkbh1 conditional knockout (cKO) mice inbred C57BL/6 background. These showed reduced respiratory chain complex, atrophy, CA1 pyramidal neuron abnormalities. Furthermore, performances fear-conditioning Morris water maze tests cKO indicated that abnormalities led impaired hippocampus-dependent learning. findings indicate critical roles hippocampus.

Kv4.2 knockout mice have hippocampal-dependent learning and memory deficits.
Kv4.2 channels contribute to the transient, outward K(+) current (A-type current) in hippocampal dendrites, and modulation of this current substantially alters dendritic excitability. Using Kv4.2 knockout (KO) mice, we examined the role of Kv4.2 in hippocampal-dependent learning and memory. We found that Kv4.2 KO mice showed a deficit in the learning phase of the Morris water maze (MWM) and sig...

Plasticity in dendritic spines may underlie learning and memory. Spinophilin, a protein enriched in dendritic spines, has the properties of a scaffolding protein and is believed to regulate actin cytoskeletal dynamics affecting dendritic spine morphology. It also binds protein phosphatase-1 (PP-1), an enzyme that regulates dendritic spine physiology. In this study, we tested the role of spinoph...
متن کاملCB1 knockout mice display impaired functionality of 5-HT1A and 5-HT2A/C receptors.
Interaction between brain endocannabinoid (EC) and serotonin (5-HT) systems was investigated by examining 5-HT-dependent behavioral and biochemical responses in CB(1) receptor knockout mice. CB(1) knockout animals exhibited a significant reduction in the induction of head twitches and paw tremor by the 5-HT(2A/C) receptor selective agonist (+/-) DOI, as well as a reduced hypothermic response fo...
متن کاملGlucagon receptor knockout mice display increased insulin sensitivity and impaired beta-cell function.
In previous studies, glucagon receptor knockout mice (Gcgr(-/-)) display reduced blood glucose and increased glucose tolerance, with hyperglucagonemia and increased levels of glucagon-like peptide (GLP)-1. However, the role of glucagon receptor signaling for the regulation of islet function and insulin sensitivity is unknown. We therefore explored beta-cell function and insulin sensitivity in G...
متن کاملImpaired Terminal Differentiation of Hippocampal Granule Neurons and Defective Contextual Memory in PC3/Tis21 Knockout Mice
Neurogenesis in the dentate gyrus of the adult hippocampus has been implicated in neural plasticity and memory, but the molecular mechanisms controlling the proliferation and differentiation of newborn neurons and their integration into the synaptic circuitry are still largely unknown. To investigate this issue, we have analyzed the adult hippocampal neurogenesis in a PC3/Tis21-null mouse model...
